Argonaute-mediated Cleavage Arrow Ontology Artificial Evolution
نویسنده
چکیده
Artificial evolution refers to any procedure that uses the mechanism of Darwinian evolution to generate a product. While this definition encompasses the evolution of organic life forms by means of artificial selection (breeding of animals, plants, or microorganisms), artificial evolution commonly refers to the instantiation of evolution within a nonbiological medium. Artificial evolution is sometimes used synonymously with ▶ artificial life, but in principle the concepts are overlapping but different.

Crystal structure of A. aeolicus argonaute, a site-specific DNA-guided endoribonuclease, provides insights into RISC-mediated mRNA cleavage.
Argonaute (Ago) proteins constitute a key component of the RNA-induced silencing complex (RISC). We report the crystal structure of Aquifex aeolicus Ago (Aa-Ago) together with binding and cleavage studies, which establish this eubacterial Ago as a bona fide guide DNA strand-mediated site-specific RNA endonuclease. We have generated a stereochemically robust model of the complex, where the guide...

MicroRNAs (miRNAs) play an important role in targeted gene silencing by facilitating posttranscriptional and translational repression. However, the precise mechanism of mammalian miRNA-mediated gene silencing remains to be elucidated. Here, we used a stem-loop array reverse-transcription polymerase chain reaction assay to analyse miRNA-induced mRNA recognition, cleavage, posttranscriptional mod...
متن کاملStructure/cleavage-based insights into helical perturbations at bulge sites within T. thermophilus Argonaute silencing complexes
We have undertaken a systematic structural study of Thermus thermophilus Argonaute (TtAgo) ternary complexes containing single-base bulges positioned either within the seed segment of the guide or target strands and at the cleavage site. Our studies establish that single-base bulges 7T8, 5A6 and 4A5 on the guide strand are stacked-into the duplex, with conformational changes localized to the bu...
متن کاملMicroRNA-directed cleavage of targets: mechanism and experimental approaches
MicroRNAs (miRNAs) are a large family of post-transcriptional regulators, which are 21-24 nt in length and play a role in a wide variety of biological processes in eukaryotes. The past few years have seen rapid progress in our understanding of miRNA biogenesis and the mechanism of action, which commonly entails a combination of target degradation and translational repression. The target degrada...
متن کاملExpression determinants of mammalian argonaute proteins in mediating gene silencing
RNA interference occurs by two main processes: mRNA site-specific cleavage and non-cleavage-based mRNA degradation or translational repression. Site-specific cleavage is carried out by argonaute-2 (Ago2), while all four mammalian argonaute proteins (Ago1-Ago4) can carry out non-cleavage-mediated inhibition, suggesting that Ago1, Ago3 and Ago4 may have similar but potentially redundant functions...
